Validation and QC Services

The UConn Stem Cell Core offers validation and quality control (QC) services applicable to human embryonic stem (ES) cells and human induced pluripotent stem (iPS) cells.  They may be useful for initial derivation and characterization, prior to experiments, supplementation to publication submissions, as well as for general tissue culture lab QC at regular intervals.

These validation and QC services and reagent are available from the UConn Stem Cell Core:

Mycoplasma testing: The UConn Stem Cell Core mycoplasma testing service is performed using the Myco-Alert kit from Lonza.  Mycoalert kit FAQ

For more information about mycoplasmal biology and importance, see Bionique Laboratories excellent FAQ: (Note: Bionique is a commercial testing service.)

 Immunostaining kit: to detect pluripotency markers including cell surface markers, e.g., SSEA-3, SSEA-4, TRA1-60, and TRA1-81, and intracellular markers, e.g., NANOG if no exogenous NANOG was introduced to the cells for reprogramming.  The Stem Cell Core will provide you with aliquots of the antibodies that you need, as well as protocols.

 Gene expression by RT-PCR kit: to determine expression of endogenous OCT4, NANOG, SOX2, LIN28, KLF4, and c-MYC.  The Stem Cell Core will provide you with aliquots of the primers that you need, as well as protocols.

 Teratoma formation: to test pluripotency in vivo:  cells are injected into Immunity-deficient mice.  Any tumors that form are analyzed to look for tissues representing all three germ layers.  The Stem Cell Core will perform this service for you.
